Abstract
The invention provides human gene ABCA2-encoding nucleic acids (including cDNA, DNA and mRNA mols.), the protein encoded thereby, and antibodies specific for said protein. Based on sequence homol. and protein motif characterization, the invention claims ABCA2 as an ortholog of mouse ABCA2. The invention also provides the cDNA sequence, as well as corresponding amino acid sequence of human ABCA2. The invention further provides an expression vector (plasmid or retroviral vector) contg. said cDNA sequence, and a host cell transformed with said vector, which can be used for the recombinant prodn. of ABCA2. Still further, the invention provides transgenic knockout mice, wherein the ABCA2 gene was inactivated, and a method for screening a compd. for the ability to inhibit human ABCA2-mediated transporter activity. The invention relates that inhibition of human ABCA2-mediated transport is indicated by restoration of anticancer drug sensitivity, and/or by a redn. of transporter mediated cellular efflux of anticancer agents. Finally, the invention provides a kit for detecting the presence of human ABCA2-encoding nucleic acids, which contains a primer specific for ABCA2, polymerase, buffer and control DNA. The invention related that knockout mice will provide a valuable biol. system for assessing resistance to chemotherapy in an in vivo tumor model. In the examples, the invention demonstrated human ABCA2 gene mRNA is expressed in the highest amt. in brain. [on SciFinder (R)]
